Randolph “Randy” Murdaugh III was born in Savannah, Chatham County, Georgia, United States to Randolph “Buster” Murdaugh Jr. and Gladys Willis Marvin Murdaugh. From 1940 to 1986, Buster was the solicitor for the 14th Judicial Circuit, a division of the South Carolina judicial system.
When Randy was young, he would accompany his father to work and grew to love the courts. Randy himself became the solicitor for the 14th Judicial Circuit and tried more than 200 murder cases.

- In 1957, he graduated from Wade Hampton High School in Varnville, Hampton County, South Carolina, USA where he was very active in sports. He and Elizabeth “Libby” Alexander Murdaugh also started dating there. He was later inducted into the Wade Hampton Athletic Hall of Fame.
- In 1961, he graduated from the University of South Carolina in Columbia, South Carolina with a bachelor’s degree in business administration. In the same year, he married Libby.
- In 1963, Libby gave birth to their daughter Lynn Murdaugh, who became Allen Goettee‘s wife.
- In January 1964, he graduated from the University of South Carolina School of Law in Columbia. After his graduation, he, Libby and Lynn moved to Varnville, Hampton County and practiced law with his father in their family firm his grandfather Randolph Murdaugh Sr. founded in 1910. - In 1966, Libby gave birth to their son Randolph “Randy” Murdaugh IV.

- On December 31, 2005, he retired as solicitor for the 14th Judicial Circuit. In 2006, he rejoined the law firm Peters, Murdaugh, Parker, Eltzroth & Detrick, P.A., which was formerly called Murdaugh, Eltzroth & Peters.
- On September 20, 2018, he received South Carolina’s highest civilian honor, the Order of the Palmetto, from South Carolina’s 117th governor Henry McMaster.
- On June 10, 2021, he died in Varnville, Hampton County, South Carolina at age 81.
